For the majority of the mounts, the answer is no. The ASIAIR will (I should say "should") do the 60 degree slew automatically for you between the initial plate solve and the subsequent ones.
Unfortunately, there are apparently rampant bugs in the 1.5.x versions of ASIAIR regarding the SynScan protocol (Celestrons predominately, but other SynScan based mounts too) that prevents that from happening. Some of the mounts simply don't slew the 60 degrees when it is supposed to (or it slews it in the wrong direction).
If mount is not able to slew by itself, you will need to do it manually by...
... first aborting the polar alignment process, and disconnect all mounts from the Telescope Setup window -- select ST4 mount, for example.
When no mount is connected, the polar alignment process will ask you to move the mount manually -- that is where you de-clutch the RA axis, rotate it 60 degrees (counterclockwise, if the mount is between you and the north celestial pole), and engage the clutch again. Be sure not to move the Declination axis (keep it locked) when you do this, otherwise the accuracy is non existent. You can also use the hand controller's East-West button to move the mount by 60 degrees. When you are done, tap the button that asks you to confirm that you have made the approximately 60 degrees (4 hours in Hour Angle) rotation.
Once you are done with polar alignment, you can then connect your mount back again in the Telescope Setup window.
If you have a mount that should slew automatically but does not, it may need to be properly star aligned before connecting to ASIAIR. Then carefully check that the hand controller's time, UTC offset and longitude are exactly the same as the numbers in the Telescope Setup window.
Before entering the polar alignment process, take a Preview image and plate solve it. Make sure plate solving is reliable -- you may need to change exposure times to make that happen, especially when the skies are not fully dark yet. Then use the same exposure time in the polar alignment window. This applies to both the mounts that auto slew and mounts that need to be manually slewed.
It is also best to home the mount before reconnecting it to ASIAIR. If your mount will auto-60 degree slew, disconnect it, home the mount with a hand controller, and then reconnect to Telescope Setup. Some people report that ASIAIR apparently loses its knowledge of some mount's coordinates when it polar aligns. I home my RainbowAstro mounts after polar alignment even though I have never seen the problem with them, albeit, I don't disconnect first -- simply home it using the home function in the ASIAIR Telescope Setup window.
Stand right by your mount and be ready to pull power in case it goes berserk after polar alignment (many people have driven the OTA train into their piers), that is why I recommend homing it once so that there is less likelihood that ASIAIR has the wrong mount coordinates after polar aligning.
